Do you have to ask for them? Are they just part of the normal reward system like food?
I travel to LV 2-3 times a year and spend time at poker, and craps/bj tables. I have all the reward cards but never really paid attention to what I earned.
I typically play at Venetian, MGM , casino royale (craps), mirage and TI.
Playing just poker you are not likely to get a room comped.
Those that spread their play out into the casino, where every dollar you lose is going directly to the house, have a better chance of getting a room comped.
The best way to find out what is possible is to ask in several locations - the poker room itself to find out if it is possible through just poker (it is not at The Mirage), at the casino's players club desk, and a slot or table games host (if you are playing slots or table games).
However, most (if not all) poker rooms do have the ability to offer their casino's Casino Rate to poker players. Some rooms have fixed amounts, others (like ours) have a fluctuating rate that varies with the hotel rate. For most rooms you will only have to average a 5 or 6 hours per day of play to qualify for the casino rate.
When it comes to getting the best rate it is always worthwhile to check the propertery's web site to see what type of hotel deals are being offered as well.

Any help?
Do the following, right now:
Pull out your Harrah's/Caesars 'Total Rewards' card;
Go online to Caesars.com;
Create a PIN & register your TR account online, if you have not already done so;
Once you are logged in with your account number, check two places, under both the "My Offers" page and also separately on the room rate calendar through the "Reservations" section of their site.
Even if you have never spent a nickel in any of their joints, you could be shocked (in a good way) to discover what they might do to entice you into their only slightly tacky dens of middle brow Wonderbread iniquity. Or not, in which case it will have taken up about ten minutes of your time. In this one department, and only this, that company is still decades ahead of the competition. Frankly I would probably never play in any of the CET rooms if not for their Total Rewards system that has got me very effectively roped into giving them about a third of my play so I can maintain the room rates (often fully comped) and other perks I get from their system. I'm not a pit or a slot player, just poker and some four-hooved type sporting events, and I discovered comps and offers waiting for me to reel me in after a few years of not putting a single point on that card. They are very good at this. I think it might be the only thing they do better than their competition, but it is a big one.
Go do it. You probably have at least two nights waiting for you that you don't even know about, maybe more, without even having to make a phone call. Then go ahead and play some poker at the Mirage, while you're in the neighborhood.
Nobody else comps me skeet for rooms. I certainly never find any reason to bother logging on to the MGM site with my player account; their hotel resort fee add-ons alone are more than the total room rate I pay across the street. The others do the usual food comp thing and have some nice casinos and hotels and some of them are superior poker rooms, but squadoosh for room comps for me. Maybe you can do better since you are also out in the pit shooting dice and stuff, but I don't know how to go about getting that, if it can be got.

I did the total rewards thing. When looking for a room it asked if there was a companion's card. I entered my wife's information in and between the two of us we are getting the entire trip comped at caesars over labor day weekend.

I'm with Rock. Ceaser's/Harrah's is the only one that ever has comped me rooms. I've gotten rooms in four different casino towns (about 4 trips/year), and rarely pay for a night (maybe 20-50 bucks on weekends). That's without asking for the poker room rate.
I usually stay at Bally's on comps. I got most of my last week long trip room nights comped having to only pay a reduced rate for the weekend. I play mostly poker but make sure to play a little of other things as well to rack up a few points on other games. I eat at the restaurants and make sure my players card is presented and I get points for that. Ceaser's also offers a Total Rewards Visa card that adds points to your account. I use the card to make sure my points do not expire. Also using the card at the properties offers extra points.
I did the reward card thing with Boyd's Gaming and got a room comped this Friday and Saturday (!!??) at the Orleans. Kind of a shock because my wife plays minimal ($100 a day) and I'm about a $500 a day gambler, mostly poker but do play a little slots and too much craps.
Use the card whenever you gamble and you will get all kinds of offers on the internet for future stays.
in the past 6 months ive gotten free rooms at Tropicana, NYNY and RIO. I even have the 1st thru the 3rd of july for free at NYNY with two complimentary tickets for the new kids on the block and back street boys concert, if was a 12 year old girl I would probably go, but im not, but the offer is still there for me.
and the free rooms at tropicana are actually free rooms, they did not make me pay the $9.99 a day fee for the stuff you never use, i was actually kind of shocked when i checked out, but on their free room offers its actually a free room, the NYNY comped rooms still cost you $14.99 a day plus tax, but thats still a really good deal IMO.
I'm sure the excalibur will send me some free rooms, im sure they want their $5,000 back that I won playing quarter slots, it was unreal considering i never win more than a hundy or so at a time on the devils machines.

This is good advice. We always have a free room coming for a Harrah's property and seldom gamble there.
